I just can't get enough of the one in 22 Jump Street (2014) - to the point that I regularly revisit reaction compilations to that specific scene online.
It's one of those "you had to be there in the theater to feel how hilarious and surprising it really was... everyone busted out laughing."
So I'm wondering if there are other scenes along the same lines.
This was the first thing I thought of. I've rarely been surprised more while watching a movie than I was when I first saw that scene.

I always bring this up in relation to that scene, but I saw in in theatres and went in blind.
When that moment happened, the whole theater was dead silent for a few moments... THEN a wave of laughter started up as soon as it kind of sunk in.

"Sex Drive" was a dumb and raunchy teen sex comedy that came out in 2008. It's about an awkward teenager who has a really hot online girlfriend who lives a few states away. She tells him that if she drives out to meet her, they could hook up. So he and his friends borrow his brother's car to drive to her.
Now, you'd think the twist is that she's not that hot, is actually a guy, or just doesn't show up. But it's funnier than that. She actually is hot just like her photos. However >!she was only flirting with him so she could steal his car. She actually does this to a few guys at a time and her accomplish is also really funny!<.

Everytime I see this movie mentioned i feel the need to bring up the show which is surprisingly great. The two main characters are extremely similar looking to young tarantino / Clooney in the movie, and the show just felt like a perfect extension to the movie (season 1 is basically the movie and then s2 / 3 continue the story).
It was even developed / directed by Robert Rodriguez and I feel like no one's aware it even exists.

But on a rewatch you realize that it was nonsensical even for the WTF stuff, and should have been caught by all those ‘Worlds Greatest Detectives’.

The Marple series had a twist added to the film lead to a pretty funny gag in the adaptation of the novel "The Body In The Library", where Miss Marple investigates the murder of a stranger dumped out of the blue in the manor of her friend Dolly Bantry.
Not even counting the original twist in the novel that the corpse ended up in Gossington Hall because >!the person who was being set up as the murderer's patsy came home late one evening, found a murdered girl in his living room and moved the corpse because he considered doing so nothing more than a prank (he was *very* drunk at the time) and this is what foiled the murder scheme!<, but Miss Marple leaves the chief inspector completely catatonic when explaining the case that >!unlike the original novel where the plot twist was that two characters were secretly married, here the murderers are a lesbian couple!<. It is fantastic.
