Timing most often gets brought up when talking about live performance, but sometimes something catches me perfectly in a movie and I always love when it does.
I was re-watching The Jerk for the first time in decades and Bernadette Peters' cornet out of nowhere strikes me as a perfectly timed joke.
There is also a tiny bit from William H. Macy in State and Main that I love. Someone says in reference to Alec Baldwin's character "What does he like?" and the response is "14 year old girls". Macy says "Well, get him something else. Get him half a 28 year old girl." and then there is a brief pause before he continues, quietly, off camera with "How's my math?". The time that passes is just long enough for the audience member to do the quick math. It is an almost surreal joke.

Honestly, in real life the heroes and villains of the movie would be reversed. "Dickless here" is a diligent government employee trying to stop a group of washouts who claim to see ghosts from running an unlicensed nuclear device. Our heroes are said washouts. The only thing that makes the movie work is that the washouts are just so damned likeable and the guy trying to stop New York from being destroyed by a nuclear accident is kinda a prick about it.

I love when facial expressions are timed perfectly. Sometimes it’s almost the actor breaking the fourth wall and looking in to the camera. There’s a scene in Secondhand Lions where the guy is beating up Haley Joel Osmont and the lion starts sprinting at him and he briefly sees it, and he turns and pauses in disbelief and looks directly at the camera before he’s attacked. Another is Eddie Murphy in Trading Places who hears one of the brothers explaining that bacon might be found on a bacon, lettuce and tomato sandwich, and Eddie just looks at the camera with a blank stare.

In the fifth element, the absolutely perfect timing of Dallas entering the elevator and Zorg exiting the one right next to it just as they disappear from view creates a hilarious anticlimax as they dodge a confrontation that had been foreshadowed for like fifteen minutes by that point.

For some reason everyone messes up quoting this. You need the full exchange:
Ray: "Everything was fine until the power was cut off by dickless here."
Peck: "They caused an explosion!"
Mayor: "Is this true?"
Venkman: "Yes it's true. This man has no d***."
The reason it's so funny is that the mayor's line is ambiguous; it's unclear if he's referring to Ray's assertion or Peck's, and he's not looking directly at either of him. So the humor comes when Venkman pauses after "Yes it's true." You either expect him to say, "Yes it's true that everything was fine until the power was shut off," or "Yes it's true that an explosion happened," and he says something completely different.
